**Background**
Mitochondrial dynamics, specifically the balance between fusion and fission, are critical for maintaining cellular homeostasis and neuronal survival. Dynamin-related protein 1 (Drp1) is a key GTPase that mediates mitochondrial fission by translocating from the cytosol to the mitochondrial outer membrane. Aberrant Drp1 activation and excessive mitochondrial fragmentation are hallmarks of various pathological conditions, including Parkinson’s disease, Alzheimer’s disease, Huntington’s disease, and cerebral ischemic injury. In these conditions, Drp1-mediated mitochondrial dysfunction often leads to oxidative stress and programmed cell death. Consequently, targeting the translocation and activation of Drp1 has emerged as a promising therapeutic strategy to preserve mitochondrial integrity and provide neuroprotection. In this context, we will introduce a selective Drp1 inhibitor – Drp1 peptide inhibitor P110.
**Definition**
Drp1 peptide inhibitor P110 is a selective peptide inhibitor designed to block the activation of Drp1 and prevent its translocation to the mitochondria.
**In Vitro and In Vivo Studies**
The Drp1 peptide inhibitor P110 description highlights its sequence as YGRKKRRQRRRGGDLLPRGS-NH2, with a molecular weight of 2411.78 (free base). Regarding Drp1 peptide inhibitor P110 biological activity, the compound acts by inhibiting the recruitment of Drp1 to the mitochondria, thereby preventing excessive fission. In vivo studies using an MPTP-induced model of Parkinson’s disease demonstrated that P110 can prevent MPTP-induced Drp1 mitochondrial translocation. Furthermore, Drp1 peptide inhibitor P110 in vivo administration effectively alleviated the loss of dopaminergic neurons, reduced damage to dopaminergic nerve terminals, and mitigated associated behavioral deficits. Beyond Parkinson’s disease, P110 has shown the ability to reduce mitochondrial damage and organ injury in animal models of Huntington’s disease, cerebral ischemic injury, and myocardial infarction. These results indicate that the peptide is a potent tool for studying mitochondrial-mediated cell death. In conclusion, Drp1 peptide inhibitor P110 is a selective inhibitor that provides significant neural protection by modulating mitochondrial dynamics.
